Lung lamellar bodies maintain an acidic internal pH.
The internal pH of lung lamellar bodies was investigated with membrane permeable basic amines. Isolated granular pneumocytes and isolated lung lamellar bodies exhibited fluorescence when exposed to 8 microM quinacrine, suggesting accumulation of this dye due to an acidic internal pH. Uptake of [14C]methylamine by isolated lung lamellar bodies was measured to quantitate the intralamellar body pH...
متن کاملYolk platelets in Xenopus oocytes maintain an acidic internal pH which may be essential for sodium accumulation
Yolk platelets constitute an embryonic endocytic compartment that stores maternally synthesized nutrients. The pH of Xenopus yolk platelets, measured by photometry on whole oocytes which had endocytosed FITC-vitellogenin, was found to be acidic (around pH 5.6). Experiments on digitonin-permeabilized oocytes showed that acidification was due to the activity of an NEM- and bafilomycin A1-sensitiv...
متن کاملSurfactant protein composition of lamellar bodies isolated from rat lung.
Lamellar bodies isolated from rat lung contain all three classes of surfactant proteins, SP-A, SP-B and SP-C, as determined by immunoblot analysis. The amounts of the surfactant proteins present in lamellar bodies, determined by sandwich e.l.i.s.a. (SP-A) and fluorescamine assay (SP-B and SP-C) show that these organelles are highly enriched in the hydrophobic surfactant proteins SP-B and SP-C.
متن کاملFormation of lung surfactant films from intact lamellar bodies.
Lamellar bodies, an intracellular source of lung alveolar surfactant, were isolated from rat lung homogenates and studied in the Langmuir-Adams surface balance. By layering intact lamellar bodies on the surface of a more dense sucrose subphase, we studied the factors affecting film formation from surface tension-vs-time data and determined surface tension-surface area isotherms by compression a...
متن کاملParasitophorous vacuoles of Leishmania amazonensis-infected macrophages maintain an acidic pH.
Leishmania amastigotes are intracellular protozoan parasites of mononuclear phagocytes which reside within parasitophorous vacuoles of phagolysosomal origin. The pH of these compartments was studied with the aim of elucidating strategies used by these microorganisms to evade the microbicidal mechanisms of their host cells. For this purpose, rat bone marrow-derived macrophages were infected with...
